DOVE
Drotaverine (INN, also known as drotaverine) is an antispasmodic drug, structurally related to papaverine. Drotaverine is a selective inhibitor of phosphodiesterase-4, and has no anticholinergic effects. Drotaverine has been shown to possess dose-dependent analgesic effects in animal models.

Mechanism of Action
- Cyclic AMP (cAMP) is a second messenger to regulate a large number of key processes.
- Phosphodiesterase (PDE) hydrolyses cAMP, thus decreasing its concentration in the cells.
- cAMP and Ca++ imbalance results in smooth muscle spasm and pain.
- Drotaverine has antispasmodic effect mediated via inhibition of phosphodiesterase-IV, specific for smooth muscle.
- Drotaverine acts to correct cyclic AMP and Ca imbalance at the spastic site, thereby relieving smooth muscle spasm and pain.
- It has a rapid and direct action on the smooth muscle.
Indications
- Intestinal and Biliary renal colics*
- Irritable Bowel Syndrome(IBS)
- Uterine Spasm
Dosage
Route of administration: Oral
Children more than 6 years: 40 mg orally 3 times a day tid.
