Drotaverine (INN, also known as drotaverine) is an antispasmodic drug, structurally related to papaverine. Drotaverine is a selective inhibitor of phosphodiesterase-4, and has no anticholinergic effects. Drotaverine has been shown to possess dose-dependent analgesic effects in animal models.

Mechanism of Action

  • Cyclic AMP (cAMP) is a second messenger to regulate a large number of key processes.
  • Phosphodiesterase (PDE) hydrolyses cAMP, thus decreasing its concentration in the cells.
  • cAMP and Ca++ imbalance results in smooth muscle spasm and pain.
  • Drotaverine has antispasmodic effect mediated via inhibition of phosphodiesterase-IV, specific for smooth muscle.
  • Drotaverine acts to correct cyclic AMP and Ca imbalance at the spastic site, thereby relieving smooth muscle spasm and pain.
  • It has a rapid and direct action on the smooth muscle.

  • Intestinal and Biliary renal colics*
  • Irritable Bowel Syndrome(IBS)
  • Uterine Spasm

 

            Dosage

Route of administration: Oral

Children more than 6 years 40 mg orally 3 times a day tid.

Contraindications

DOVE is contraindicated in the following conditions:

  • Cardiac dysfunction
  • Renal insufficiency
  • Hepatic insufficiency
